Dasatinib is an oral potent oncokinase inhibitor that can inhibit multiple oncokinases such as BCR-ABL kinase and SRC family kinases. Clinically, it is used for the treatment of Philadelphia chromosome-positive (Ph+) chronic myeloid leukemia (CML) and Philadelphia chromosome-positive (Ph+) acute lymphoblastic leukemia (ALL) in patients who are resistant to or intolerant of imatinib mesylate.In addition, some compounds prepared based on 2-amino-N-(2-chloro-6-methylphenyl)thiazole-5-carboxamide have been shown in studies to possess excellent ABL kinase inhibitory activity and antineoplastic activity. These compounds can be further developed into novel antineoplastic drugs for the treatment of diseases caused by tyrosine kinase dysfunction, such as chronic myeloid leukemia (CML), gastric cancer, and gastrointestinal stromal tumors (GISTs). |
